چکیده
Enzymes are the biocatalysts synthesized by living cells. They are Complex protein molecules that bring about chemical reactions concerned with life. They are protein in nature, colloidal and thermolabile in character, and specific in their action. L-asparaginase (L-asparagine amido hydrolase, E.C.3.5.1.1) is an extra cellular enzyme that has received considerable attention since it is used as an anticancer agent. The present work deals with production of extracellular L-asparaginase from Aspergillus terreus MTCC 1782 using Bajra seed flour under solid state fermentation Process parameters like Incubation time(96 h), Temperature (30 C), Moisture content (70% v/w), pH of the medium(8.0), Inoculum Age (5 days), Inoculum volume (1 ml), carbon source (1.5% w/v glucose), nitrogen source ( 2% w/v ammonium sulphate), and metal salts ( 0.1% w/v Magnesium sulphate) were optimized and giving an overall yield of 273.3 U/gds of maximum L-asparaginase activity after optimization. The observation made in this study hold great promise for scale up production of L-asparaginase from Aspergillus terreus MTCC 1782 using Bajra seed flour as substrate under solid state fermentation.

متن کاملOPTIMIZATION OF PROCESS PARAMETERS FOR L- ASPARAGINASE PRODUCTION BY Aspergillus terreus MTCC 1782 UNDER SOLID STATE FERMENTATION USING MIXED SUBSTRATE
L-asparaginase (L-asparagine amido hydrolase, E.C.3.5.1.1) is an extra cellular enzyme that has received considerable attention since it is used as an anticancer agent. L-asparaginase belongs to an amidase group that hydrolyses the amide bond in Lasparagine to aspartic acid and ammonia. The clinical action of this enzyme as an anti-carcinogenic is attributed to the reduction of L-asparagine; tu...
متن کاملEvaluation and Screening of Nitrogen Source for L-asparaginase Production by Aspergillus terreus MTCC 1782 using Latin Square Design
The manufacture of enzymes, like L-asparaginase with less adverse effects in the treatment of cancer is an important facet of pharmaceutical industry. Yeast and Filamentous fungi are commonly reported in scientific literature to produce L-asparaginase with less adverse effects than prokaryotic microorganisms. In the present study Latin square design was used to examine the effect of L-proline, ...
متن کاملAmylase production from Aspergillus oryzae LS1 by solid-state fermentation and its use for the hydrolysis of wheat flour
Nine Aspergillus and three of Trichoderma strains were grown on wheat bran (WB) medium under solid state fermentation (SSF) for amylase production. Aspergillus oryzae LS1 produced the highest level of the enzyme. The thermal stability profile of its crude enzyme revealed the half-life time of more than 2 h at 50 and 60ºC. The enzyme production was affected by strain type, incubation periods, le...

In this study, Glucoamylase production by Aspergillus Niger was investigated under solid state conditions with low cost by-products of agricultural processes as substrate. Highest enzyme production was observed when a combination of wheat bran (WB) and corn flour (CF) was used as compared to WB+ rice bran, WB+ rice flour and WB alone. Different additions of (CF) were tested and WB+ 10% CF showe...
